Suspend 70.3 g of the dehydrated mediumin 1,000 mL of dstilled water, mix well andheat to dissolve the medium. Sterilize byautoclaving at 121C for 15 minutes, andkeep the medium at about 55C. Distribute15 mL of medium into the anaerobicculture pouch in which 10 mL of samplehas distributed. Mix well, and weld to sealthe neck of the pouch after removing airbubbles. Incubate at 37t1’C for 24t2hours.
The medium is used for the colony countingof Clostridia (Spore forming, sulfurous acidreducing anaerobic bacterium) in foodstuff.Clostridia develop colonies with brown to black color by reducing sulfurous acid, whileothers develop light white colonies.
